I'm guessing this is an anime or homestuck-esque comic.
First you probably want to establish if this is a drama, dramedy, or comedy. Is it a day-to-day comic (most likely a comedy), or is there an overarching story (most likely dramedy or drama)? If it's a drama or dramedy, what's the main conflict of the story? If it's a comedy, what kind of comedy are you shooting for? Also knowing the art style is a huge thing. Even if you're not too great at drawing, there's still plenty you can do with a comic to make it entertaining. Going to what Sig said, please don't make this a self-insertion comic, so that means no Mary Sues.
I expect a presentation of this project and both your physical and digital portfolios, resume, and cover letter by noon tomorrow.
